Özet

This

study investigates the extraction yields, phenolic and flavonoid contents,

antioxidant capacities, and potential antidiabetic effects of various solvent

extracts from Datura innoxia Mill. seeds. The extraction yields revealed

that hexane (HEX), ethyl acetate (ETA), and methanol (MET) extracts had the

highest yield, while butanol (BUT) and aqueous (AE) extracts had lower yield.

Phenolic content was highest in the ETA and AE extracts, while the lowest was

observed in the HEX extract. In contrast, flavonoid content increased with

solvent polarity, with AE exhibiting the highest content. Antioxidant capacity

revealed that the AE extract exhibited high ABTS.+ radical

scavenging activity, while MET and HEX extracts also displayed significant

activity. DPPH. scavenging activity was strongest in the AE extract and showed a dose-dependent

increase in the HEX and ETA extracts. Superoxide radical scavenging activity was

primarily found in the MET, HEX, and AE extracts. Additionally, the extracts

displayed potent ferric reducing antioxidant power (FRAP), especially the MET,

AE, and BUT extracts. The antidiabetic potential of the extracts was evaluated

in an animal model. While higher doses of extracts did not significantly affect

blood glucose levels, 50 mg/mL AE and MET extracts demonstrated a temporary

decrease in plasma glucose. The extracts also appeared to have a positive

effect on diabetic rats' survival. It was suggested that MET, BUT, and AE extracts

obtained from D. innoxia seeds exhibited strong antioxidant properties,

with the MET extract, in particular, able to alleviate hyperglycemic effects in

the STZ-induced diabetic rats.
