Özet

Cellulose, which is mostly produced by plants in nature and forms a large part of the cell

structure of plants, is a linear polymer formed as a result of the bonding of glucose molecules

with ꞵ-1,4 glucan bonds. Cellulose is produced together with lignin and hemicellulose in

plants. In order for cellulose to be used industrially, it must be separated from lignin and

hemicellulose.

Cellulose is widely used in textile, paint, medicine, paper industry. Cellulase enzyme is

used to break down cellulose. The cellulase enzyme hydrolyzes ꞵ-1,4 glucan bonds between

the glucoses making up the cellulose. During this reaction, the conversion of cellulose to

glucose occurs with the combined operation of three enzyme groups (endoglucanase,

exoglucanase, ꞵ-glucosidase). Cellulases, which are biotechnologically significant, are also

used in the fields of food, paper, beer, wine, feed, textile and cleaning. The most important

disadvantages of cellulase, which is widely used industrially, are the expensiveness of

commercially available cellulase and the high cost of production. In this regard, many studies

are carried out to reduce the cost. One of the studies is the use of agricultural waste materials

as a carbon source.

In this study, it was aimed to produce cellulase enzyme from SHO60 isolated from clay

and pineapple peel which would be ragarded as waste material. Production parameters such as

incubation times, pH range and amount of carbon source were investigated. As a result of the

studies carried out in this context, the optimum enzyme activity
