The possibility of depletion of fossil fuel resources and the associated environmental

pollution are directing world energy policies towards the development of alternative

energy resources. The demand for energy has been increasing constantly as the

consequence of evolving technology and rapidly growing world population. Cellulose

considered as an alternative form of energy has recently gained significance and

represents millions of dollars for countries that have the opportunity to obtain energy

from it. At the same time, cellulosic biomaterials are attractive since they are both cheap

and abundant. To use this important resource, its stubborn structure must be broken

down. Undoubtedly, the most attractive environment where cellulolytic enzyme is

produced will be the rumen environment so rumen bacteria are regarded as unique for

this job. Therefore, isolation, identification and characterization of cellulolytic rumen

bacteria or determination of the prokaryotic diversity of rumen environment is of great

importance for cellulase production. For this purpose, in this study, cellulase production

was carried out with Paenibacillus validus bacteria isolated from the rumen and the

culture conditions were optimized. As a result of analysis, the optimal conditions were

determined as; 2 g/L yeast exract, pH:6, 48 hours incubation time and temperature

37oC. Under optimized conditions the cellulase enzyme activity was increased

approximately 2-fold (21 U/mL).
