The Effect of Helichrysum Plicatum Extract on Gentamycin Induced Nephrotoxicity in Rats.
Özet
Abstract
In this study was aimed to investigate effects on the levels of liver and renal tissue MDA, SOD, GPx and
CAT activities of the possible protective or therapeutic effects of Helichrysum plicatum (HP) extract of
gentarnycin induced nephrotoxicity. 36 Sprague Dawley rats were used as live material, it was formed six groups
containing each 6 rat and provided its adaptation to laboratory for 7 days. 1. Group control group: %5 DMSO
(ip), 2. Group HPEl group: HPE 100 mg/kg/day/ip 8 days, 3. Group HPE2 group: HPE 200 mg/kg/day/ip 8
days, 4. Group GM (Gentamycin) group: gentamycin as 80 mg/kg/day/ip 8days, 5. Group GHPEl group:
gentamycin as 80 mg/kg/day/ip + HPE 100 mg/kg/day/ip 8 days and 6. Group GHPE2 group: gentamycin as 80
mg/kg/day/ip + HPE 200 mg/kg/day/ip 8 days. At the end of these processes, the animals were sacrificed, liver
and renal tissue were excised by dissection and the changes in the cellular antioxidant defense system were
evaluated by using biochemical methods. Liver, renal tissues were collected for histopathological evaluations.
Renal and liver tissue MDA levels were measured to determine the !eve! of oxidative stress; SOD, GPx and CAT
activities were measured. Changes in ali of the parameters analyzed statistically significance were found
between the groups (p<0.001). in GHPEl group, it was shown that the activity ofSOD, GPx and CAT activities
increased and decreased levels of MDA. HPE extracts 100 mg/kg doses of demostrated pos iti ve effect on the
nephrotoxicity rat liver and renal tissue by partially supporting an antioxidant defense in nephrotoxicity rats and
have therapeutic effects on liver and renal tissues and biochemical parameters. It was found that HPE 200 mg/kg
in significantly decreased SOD, GPx and CAT activities, increased MDA levels in nephrotoxicity rats. Both
biochemical results and histopathological evidence showed that administration of HPE 100 mg/kg dose reduced
the gentamicin-induced nephrotoxicity and hepatotoxicity. As a result, it was concluded that HPE 100 mg/kg
dose has enough therapeutic feature.
